Rob Dillingham was born Robert Deon Potasi Dillingham on January 4, 2005 in Hickory, North Carolina to Valaaulia Tailele and Donald Dillingham.  He has two siblings, Pai Tailele and Denzel Dillingham.

Dillingham dunked for the first time during the summer of his ninth-grade year in Rock Hill, South Carolina.  In high school, he was ranked as high as No. 13 overall by 247Sports.  He appeared in the 2023 Iverson Classic and was the Iverson Classic Crossover Award winner.  He was the Charlotte Observer High School Basketball Player of the Year in 2020-21 and was the 2021 FIBA Americas U16 Championship MVP, averaging 15.7 points and 6.2 assists in more than 20 minutes per game.  Dillingham averaged 21.2 points, 4.9 rebounds, and 2.1 steals per game as a sophomore at Combine Academy, leading the team to a 29-3 record and a non-association state title.  He joined Overtime Elite for his final high school season.  Dillingham averaged 14.7 points and 4.9 assists per game at Overtime Elite

Dillingham chose Kentucky over North Carolina, UCLA, Southern Cal, Memphis, and Louisville.

As a freshman at Kentucky, Dillingham was tabbed the Southeastern Conference Sixth Man of the Year and was selected to the All-SEC Second Team by both the league’s coaches and media.  He was also named to the All-SEC Freshman Team.  Dillingham earned SEC Freshman of the Week and Associated Press National Player of the Week following a career-high 35 point-performance against top-five ranked Tennessee.  He was Andy Katz’s selection for National Freshman of the Year.  Dillingham averaged 15.2 points, 3.9 assists and 1.0 steals per game and connected on 47.5% from the field and 44.4% from 3-point range.  He sank 64 3-pointers and hit double-figure scoring in 27 games, including eight games with 20 or more.  Dillingham led UK in scoring eight times and assists in 12 games.  He scored a team-high 27 points and added four rebounds and seven assists in the SEC Tournament.  He logged 17 points, five rebounds, three assists and a pair of steals in his collegiate debut against New Mexico State.

On April 9, 2024, Dillingham announced that he would be entering the 2024 NBA Draft.
